Overview

CJC-1295 DAC is a modified form of GHRH with a 'Drug Affinity Complex' that binds to blood albumin. This extends its active life from minutes to over a week. Unlike the 'No DAC' version which creates natural pulses, this version creates a continuous 'bleed' of Growth Hormone and IGF-1 for up to 6 days after a single injection.

Mechanism of Action

Functions as a Long-Acting GHRH Agonist. The key differentiator is the DAC (Drug Affinity Complex)—a lysine linker attached to a reactive maleimide group. Upon injection, this group chemically bonds to serum albumin (the most abundant protein in blood). This protective shield prevents the peptide from being broken down by enzymes (DPP-IV), extending its half-life to 6-8 days. Consequently, it keeps the pituitary gland under constant stimulation, resulting in a continuous, non-pulsatile elevation of basal GH and IGF-1 levels. This is often called the 'GH Bleed' effect.

However, modern research (2020-2024) often contrasts this with the 'No DAC' version, noting that while DAC provides convenience (once weekly dosing), the continuous elevation of GH can desensitize pituitary receptors and induce temporary insulin resistance ('GH diabetes') more readily than pulsatile peptides. It is thus often studied in short bursts for acute tissue repair rather than long-term anti-aging.
