5-Amino-1MQ
NNMT inhibitor for fat loss and NAD+ restoration
Overview
5-Amino-1MQ is a small molecule inhibitor of NNMT (nicotinamide N-methyltransferase). By inhibiting this enzyme, it prevents the waste of nicotinamide, forcing it back into the NAD+ salvage pathway. This leads to increased intracellular NAD+, higher metabolic rate, and 'shrinking' of white adipose tissue. It is a key compound in longevity research for mimicking the metabolic benefits of calorie restriction.

Mechanism of Action
Functions as a selective, membrane-permeable inhibitor of NNMT (Nicotinamide N-methyltransferase). Normally, NNMT methylates nicotinamide (Vitamin B3) into 1-MNA, removing it from the NAD+ cycle and promoting fat storage. By blocking NNMT, 5-Amino-1MQ forces nicotinamide into the NAD+ salvage pathway, significantly boosting intracellular NAD+ levels. This rise in NAD+ activates Sirtuin-1 (SIRT1), the 'longevity gene,' which enhances mitochondrial oxidative phosphorylation and promotes the browning of white fat (increasing energy expenditure). The result is a dual effect: improved cellular energy (ATP) and reduced lipogenesis (fat accumulation).

Recent Research
Current research (2024–2025) positions 5-Amino-1MQ as a breakthrough in 'NAD+ therapeutics.' Unlike precursors (NR, NMN) that add fuel to the fire, 5-Amino-1MQ 'plugs the leak' by stopping the enzymatic waste of NAD+. New studies from Frontiers in Pharmacology highlight its ability to reverse muscle aging (sarcopenia) by rejuvenating muscle stem cells. Additionally, metabolic research confirms its efficacy in reducing visceral fat depots in obese models while maintaining muscle mass, a distinct advantage over GLP-1 agonists which often cause muscle loss. It is currently being explored as a dual-action agent for treating metabolic syndrome and frailty in the elderly.
